It depends on the individual hotel's policy. Most often the bouquet is handed to reception or the concierge, who notify the guest.

Many hotels don't allow couriers onto the floors for security reasons. In that case reception or the concierge accepts the flowers and either brings them to the room or gives them to the guest on request.

If in-person hand-over matters to you, add the recipient's phone number so the courier can arrange to meet them in the lobby.
